Cytoplasmic and cortical factors participating in cleavage furrow formation in eggs of three amphibian genera; Ambystoma, Xenopus and Cynops.

چکیده

A cytoplasmic factor and a cortical factor participating in cleavage furrow formation had been previously found in Cynops eggs (Sawai, 1972). These were investigated in the present study in Ambystoma and Xenopus eggs, by the method of cytoplasmic or cortical transplantation. Results were practically similar to those previously obtained with Cynops, indicating that in all the species of eggs during cleavage, the cytoplasm localizing along the cleavage furrow possessed furrow-inducing activity, and the cortex was competent to form the furrow in response to the activity of the cytoplasmic factor. In the present work, the species specificity of the two further factors was examined among Cynops, Ambystoma and Xenopus eggs, and the factors were found to act across species.

Activators of protein kinase C trigger cortical granule exocytosis, cortical contraction, and cleavage furrow formation in Xenopus laevis oocytes and eggs

Prophase I oocytes, free of follicle cells, and metaphase II eggs of the amphibian Xenopus laevis were subjected to transient treatments with the protein kinase C activators, phorbol 12-myristate 13-acetate (PMA), phorbol 12,13-didecanoate, and 1-olyeoyl-2-acetyl-sn-glycerol. In both oocytes and eggs, these treatments triggered early events of amphibian development: cortical granule exocytosis,...

A Cytoplasmic Factor Required for Contraction of the Cleavage Furrow in af Mutant Eggs of Xenopus laevis

The abnormal furrow (af) gene was identified through a maternal effect mutation that causes a defect in cell division in Xenopus embryos. In eggs obtained from mutant females (af eggs), polar body formation and cytokinesis are completely inhibited while nuclear division continues uninterrupted. Cleavage furrows are recognized as unpigmented narrow bands on the egg surface but they do not contra...

Mechanism of cleavage of newt eggs.

Cytokinesis by contraction of the base of the cleavage furrow and the dependence of the position of the future furrow on the arrangement of cytoplasm under it have been demonstrated at relatively earlier stages of cleavage in amphibian eggs. The following experiments and observations on dividing eggs of the newt Cynops pyrrhogaster indicated the presence of the contracting force at the base of ...

Roles of cortical and subcortical components in cleavage furrow formation in amphibia.

In the eggs of the newt, Triturus pyrrhogaster, 2 separate factors are recognized which take part in cleavage furrow formation. (1) The inductive capacity for the furrow formation by the cytoplasm lying under the cortex along the cleavage furrow (FIC); and (2) the reactivity of the overlying cortex to form a furrow in response to FIC. (1) FIC. The inductive capacity is shown by the fact that FI...

Calcium buffer injections inhibit cytokinesis in Xenopus eggs.

A slow cortical wave of high calcium accompanies the elongation of cleavage furrows in medaka fish eggs as well as in Xenopus eggs. We explored the role of such waves by injecting calcium buffers into Xenopus eggs at various times before and during first and second cleavage. Injection earlier than about 15 minutes before first cleavage normally starts delays it for hours. Injection between abou...
